This close collaboration fosters faster, more informed decision-making and ensures robust, high-quality data generation, ultimately increasing project success rates and delivering better outcomes for our clients. Our company is all about integrated drug discovery.
We have worked with widely different chemistry projects over the years. Two analytical chemistry service areas have emerged as RGD expert areas: Analysis & Synthesis of Peptides and Impurity Identification.
Analytical and Separation Services
Our analytical chemistry services are centered around state-of-the-art chromatographic and spectroscopic technologies, enabling us to tackle complex chemical challenges of small molecules, peptides, macrocycles, conjugates, carbohydrates, organic metal complexes, polymers and more. Our modern instrument park enables our highly skilled scientists to deliver fully customized analytical chemistry services. Our key analytical technologies are:
- Nuclear Magnetic Resonance Spectroscopy (NMR) – three spectrometers
- Mass Spectrometry (MS) – several instruments ranging from high resolution mass spectrometry to techniques suitable for low level quantification
- Liquid Chromatography (HPLC/UPLC equipped with UV, MS and/or RI detectors)
- Supercritical Fluid Chromatography (SFC) – analytical and preparative systems with mass-detection
In addition, we can generate complementary Infrared Spectroscopy (IR) data in-house.
Our analytical chemistry capabilities can be used to address a range of analytical challenges ranging from highly explorative studies to confirmatory studies. In many cases, we develop and validate analytical methods that can be used for routine quality control (QC) monitoring or transferring to other accredited QC.
Whether you need support with structural elucidation, quantification, residual solvents or identification of unknown compounds such as by-products, metabolites and contaminants, we provide innovative and robust analytical solutions tailored to your needs.
Besides our general analytical capabilities, we offer services based on our excellent PEPTIDE ANALYSIS platform. We routinely perform structure verification of synthesized products and structure elucidation of peptides and small proteins. Amino acid sequencing is performed using MS or NMR. Characterization of secondary and 3D structure of peptides and proteins is performed by 2D and 3D NMR techniques. NMR can also be used for studies of protein and peptide dynamics and aggregation behavior. We have validated NMR methods available for determination of peptide content and robust quantification of counter ions, residual solvents and small molecule impurities without the need of any prior separation.
Our advanced separation capabilities enable us to purify samples in high purity from raw products or to isolate compounds of interest from complex mixtures for further in-depth characterization by NMR or MS.
We offer both routine and customized purification and separation services based on:
- Mass-directed preparative HPLC (preparative HPLC-MS)
- Mass-directed preparative Supercritical Fluid Chromatography (SFC-MS
- Chiral separation using SFC
- Extraction, distillation and crystallization
In chemistry and pharmaceuticals, chirality is more than a structural detail — it can mean the difference between a life-saving drug and a harmful compound. Many molecules exist as enantiomers — mirror images of each other. While they may appear identical, their biological effects can be drastically different. One enantiomer might be therapeutic, while the other could be inactive or even toxic. At RG Discovery, we utilize SFC as our main method for chiral analysis and separation. As SFC uses supercritical CO₂ as the primary mobile phase, the need for organic solvents is minimized — making it a greener, more sustainable choice.
Overall, our analytical lab is designed to be agile and responsive — delivering results that are aligned with your timelines and regulatory requirements.
Advanced Synthetic Chemistry
Our synthetic chemistry teams with more than 20 chemists specialize in solving a wide range of synthetic challenges. From early-stage discovery to complex custom synthesis against specifications, we offer deep knowledge in the design and production of:
- Small molecules
- Peptides
- Macrocycles
- Carbohydrates
- Linkers and Conjugates
- Linker and “cage structure” to complex with radiometals
- Radioligand precursors
- Polymers
We apply a broad toolbox of traditional and modern synthetic methodologies, supported by computational chemistry to design efficient synthetic routes. In our well-equipped organic chemistry laboratories, the chemists routinely work with the synthesis of commercially unavailable building blocks, amino acids derivatives, intermediates, reference compounds, impurities and final compounds. We are experienced with synthesis of milligram to multigram scale deliveries of the final product.
Since the inception of RGD, we have been involved in peptide chemistry, and it has now emerged into one of our main areas of expertise. We design and perform both solution and solid phase synthesis of peptides and macrocyclic molecules, normally using automated peptide synthesizers. We take advantage of the latest developments in the peptide field and are very experienced with various chemical modifications including macrocyclization, bi- and tricyclic peptides, click chemistry, mixed disulfide bridges, ring closing metathesis, lactamization and reductive amination. Our working knowledge is also considerable regarding native chemical ligation and conjugation of peptides with PEGs, lipids, fluorescent dyes, biotin, and other conjugates.
The analytical and separation instrumentation capability facilitates the purification of peptides with up to gram scale using RP-HPLC and flash chromatography. We do regularly perform counter ion exchange of the final peptides – the most common being trifluoroacetate, hydrochloride and acetate.
